What data transfer items should I consider to estimate the cost of a inter-region VPC peering? Only Data Transfer OUT From Amazon EC2 To Another AWS Region?

Yes - the customers will be charged only for data transfer out. From in the docs: "Data transferred across Inter-Region VPC Peering connections is charged at the standard inter-region data transfer rates."
