How long does it take for AWS to approve the quota increase for Appstream users? (Basic support plan)

It takes a couple days.

User Pools max out at 50 users, and are [not recommended for production](https://docs.aws.amazon.com/whitepapers/latest/best-practices-for-deploying-amazon-appstream-2/authentication.html) environments. You should consider using SAML 2.0. There are instructions for many [SAML providers](https://docs.aws.amazon.com/appstream2/latest/developerguide/external-identity-providers-further-info.html).
