RDS proxy does not currently support custom endpoints. It goes with the default reader end point. The option would to assign a tiering to all of your reader instances and the instance with highest tiering ( lowest number) will be promoted in case of a failover.

Thanks for your reply, it helps knowing it's not there. Tiering the reader, though, doesn't prevent it from being used, does it?
I'm thinking the solution I need is to actually make a separate read-only replica cluster as described here https://docs.aws.amazon.com/AmazonRDS/latest/AuroraUserGuide/AuroraMySQL.Replication.html#AuroraMySQL.Replication.MySQL
Tiering will def not help your issue where you want connections to not go to a specific reader instance, but it can help in case of failover.
