Hello,
Websites running on an EC2 instance might become unreachable for multiple reasons. To resolve this issue, confirm that the configuration settings on your EC2 instance are correct. For example, if your instance isn't booting correctly or doesn't have the right DNS configurations, you can't connect to any website hosted on that instance.
Refer this article for the steps involving troubleshooting & open up a case with AWS Premium Support, if you are still unable to resolve the issue. There could be multiple elements involved & a live troubleshooting session would help to resolve the same quicker.

Its not VPC level issue but web service on your server is not running
if you are using apache webserver try this https://phoenixnap.com/kb/ubuntu-start-stop-restart-apache
if you are using nginx then might need to check this
But its not ACL/SecurityGroup or routing issue but server internal OS or Web service issue
