What data transfer items should I consider to estimate the cost of a inter-region VPC peering? Only Data Transfer OUT From Amazon EC2 To Another AWS Region?

Yes - the customers will be charged only for data transfer out. From in the docs: "Data transferred across Inter-Region VPC Peering connections is charged at the standard inter-region data transfer rates." Also, Inter-region VPC peering is now available globally: https://aws.amazon.com/about-aws/whats-new/2018/02/inter-region-vpc-peering-is-now-available-in-nine-additional-aws-regions/
