Can we still use the lightsail instance after being exported in the ec2 instance? or does it still work?

We want to migrate lightsail instance to ec2 instance because lightsail doesn't support path based routing.

Yes, you can migrate your instance from Lightsail to EC2. See here for the process to do so:
