Hi there!
If you're routing all traffic through the EC2 NLB, the pricing will be based on NLB rates, not Lightsail. The 100GB free tier for EC2 applies to the NLB traffic. After that, you'll be charged standard NLB data transfer rates. Traffic between the NLB and your Lightsail instance is usually free since it's internal within AWS.
