I changed the UI mode to default instead of "New EC2 Experience"(switch on the left top screen). After that "Disable source/destination" works.

I'm seeing the popup. Could be a browser issue. Try switching to another browser. Or fire up CloudShell and just issue the CLI command.

I'm seeing the same - "Actions, Networking, Change source/destination check" does nothing.

You need to select the NAT gateway and then under Details you will see Primary Network Interface ID or ENI. Clicking on the ENI will redirect you to the ENI page and from there you can click on the ENI and then the option to change Source/Destination check is available for the user to change.

Can you tell me what browser you are using? I tested on chrome(windows, mac), safari, opera(mac), firefox(mac) and it did not work at all.
Chrome on Windows.
