Disable Root Login before Marketplace Submission

According to the final checklist, it's recommended that "Root login is locked and disabled".

The AMI I am creating is basically Amazon Linux 2023 with some additional software installed. How do I lock the root account? Is it PermitRootLogin no?

Yes that's correct, although I'm pretty sure that's already the default setting with all the Amazon Linux AMIs anyway. So if you haven't changed it away from that setting, there's no need to do anything here.
