Hello.

Route53 charges fees for all but some DNS queries.  
There is also a charge for creating a hosted zone, so it depends on how you are using Route53, but there will be a charge.  
How are you using Route53 specifically?

Amazon Route 53 charges can arise even if you're within the AWS Free Tier

Hosted zones are not free
$0.50 per hosted zone per month for the first 25 hosted zones
$0.10 per hosted zone per month for additional hosted zones
