I have multi account connected via transit gateway I need to private domain to call between 2 EC2 Pleas advise design and configure

You can associate the private hosted zone to VPCs in other accounts with CLI. By association, mutual name resolution is possible.

    Note that the DNS resolution will NOT traverse the TG, it uses Route53 backend resolver to resolve domain names between Accounts. This is carried out under the covers.
