Yes, you can change the Email/password/mfa of root user without any issues.

password: https://docs.aws.amazon.com/IAM/latest/UserGuide/id_credentials_passwords_change-root.html
MFA: https://aws.amazon.com/blogs/security/reset-your-aws-root-accounts-lost-mfa-device-faster-by-using-the-aws-management-console/
Just make sure you have access to that email address and MFA device.
